Inara is teaching Mal to use a sword. They're getting intimately close to each other, and are having a "moment":
Inara: "We're fragile creatures. It takes less than a pound of pressure to cut skin.
Mal: "You know that? They teach you that at whore academy?"
This one absolutely cracked me up. Knowing Mal and Inara this was just the perfect thing to say at that time, and I had to pause the movie to laugh

whore academy heh, I remember that now that you bring it up....hilarious.
One that I remember from the movie was the whole grenades thing. Not really quotable.
